If you use blueMSX, you have 2 options :
1 - Create a dsk file with DiskManager that you'll find on the RuMSX site, drag and drop the .bas file to the window of the new dsk file in DiskManager, save the dsk file, then run blueMSX with this dsk file inserted in the virtual drive A
2 - Use the dir as dsk feature of blueMSX : create a new directory on your PC harddisk, put the .bas file in this directory, then run blueMSX with this PC directory inserted in the virtual drive A
In both cases, when you come on the blue MSX screen, you will be in MSX-BASIC. Type then RUN "filename.bas" and press on RETURN (ENTER on the PC keyboard)
